Overview

Data collected from the Northern Ireland Cohort of Longitudinal Ageing will be used to investigate genetic and epigenetic variation affecting serum antioxidants and metabolic activity. Such variation has been previously associated with poorer cognition and dementia. Improved understanding of the genetic architecture will inform future supplementation or dietary intervention trials.

Previous prospective and cross-sectional studies have identified associations between serum antioxidant levels and cognitive outcomes. Supplementation and dietary intervention have provided contrasting support for the beneficial effects antioxidants may offer in protecting against cognitive decline. Genetic variants in several metabolic pathway genes have been shown to attenuate antioxidant levels. We seek to investigate if underlying genetic and epigenetic variation influences antioxidant levels and subsequent decline in cognitive function.
Genetic data is available for 3,000 participants from the Northern Ireland Cohort of Longitudinal Ageing following genotyping using the Illumina Infinium CoreExome chip with ~550k variants and the Infinium MethylationEpic array This genome (GWAS) and epigenome wide association study (EWAS) data will be investigated against serum antioxidant measures and cognitive function measures captured a 1) baseline and 2) 10 year follow-up.
This investigation will provide improved understanding of the genetic architecture associated with serum antioxidant levels and their effect on cognitive outcomes that will inform future supplementation or dietary intervention trials.
